Our cells are regularly tested for their immunological properties to guarantee a high-quality standards for all of our vials and batches.
On the cytokine level, batches of vials are routinely tested for their secretion properties of certain key cytokines named IFN-γ, IL-2, IL-4, IL-5, and IL-17 with the ELISPOT assay.
To validate functional T-Cells, several ELISPOT assays are performed with CEF peptides from human Cytomegalovirus (CMV), Epstein-Barr Virus (EBV) and Influenza Virus. To investigate cytotoxic CD8+ T-Cell function, vials have been tested for reactivity to 32 Class-I individual CEF peptides and two Class-I CEF peptide pools. To review CD4+ T helper cell function, reactivity to 24 Class-II individual CEF peptides, one Class-II CEF peptide pool, and six Protein antigens (from Mosquito, Dust Mite, Mumps, Candida, PPD, PHA) have been tested. Additionally, cells have been tested for reactivity to the CPI-pool, a complete new developed peptide pool specifically for CD4+ T helper cells.
If you are additionally interested in a specific HLA type, our donors have been tested on high resolution HLA-typed for three Class I loci (A, B, C) and four Class II loci (DRB1, DQA1, DQB1, and DP). Moreover, we offer our customers fully characterized demographics, vaccination status and antigen reactivity corresponding to our donors. This is helpful in order to have a reference sample and designing designing further experiments.
Our ePBMC® vials are isolated from leukocytes collected by leukapharesis and are frozen freshly in CTL-Cryo™ ABC serum-free freezing medium.
The leukapharesis samples are collected in full compliance with the US HIPAA guidelines to eliminate any potential breach of privacy. To ensure that our samples are not contaminated with an infectious virus, we always serological tests of HBV surface antigen (HBsAg), Hepatitis B core antibody (HBcAb), Hepatitis C virus (HCV), Human T-Cell lymphotropic virus type 1 and type 2 (HTLV I/II) and sexual transmitted diseases (STD). Further, we used nucleid acid testing to detect a potential contamination with human immunodeficiency virus 1 (HIV I), HCV and West Nile virus (WNV).
